چکیده

The presence of endometrial tissue outside of the uterine cavity is named endometriosis and is the most common gynecologic disorder in women. Determining the inhibitory effect of a Deforolimus on angiogenesis in a three-dimensional (3-D) culture of human endometrial stromal cells (hEnCs) in vitro. The important mechanism in the pathogenesis of endometriosis is angiogenesis, and deforolimus has been shown to have anti-angiogenic activity. This was an in vitro study of human endometrial stromal cells in 3-D culture of fibrin matrix. Endometrial stromal cells isolated and placed in a 3-D fibrin matrix culture system for angiogenesis with VEGF and inhibit angiogenesis by deforolimus. Finally these cells analyzed by CD31 antibodies. After 3 weeks, in cells treated with VEGF, endothelial cell branching was observed and rudimentary capillary-like structures formed. In the presence of 5 μM of deforolimus, angiogenesis was reduced. The deforolimus were shown to be effective in inhibiting the mechanisms of angiogenesis.

Angiogenesis Following Three-Dimensional Culture of Isolated Human Endometrial Stromal Cells

Background Endometriosis is the presence of endometrial tissue outside of the uterine cavity and is the most common gynecologic disorder in women of reproductive age. We have preliminary evidence that in the presence of a 3-dimensional (3-D) fibrin matrix, human endometrial glands, stroma, and neovascularization can develop in vitro, mimicking the earliest stages of endometriosis. The aim of th...

P-96: Survey of In Vitro Effect of Noscapine on Nitric Oxide Secretion of Human Endometrial Stromal Cell

Background: Noscapine is cough suppressant drug with cell inhibitory effect. It is introduced for anti-cancer treatment. The aim of present study was to determine in vitro noscapine effect on nitric oxide (NO) secretion by human endometrial stromal cells. Materials and Methods: Human endometrial biopsies (n=8) were obtained in sterile condition and were chopped mechanically and digested by enzy...

P-99: Survey of Noscapine Effect on Apoptosis of Human Endometrial Stromal Cells: An In Vitro Study

Background: Noscapine is a water soluble alkaloid with antitussive property and very less toxicity. It is obtained from the Papaveraceae family. Recently, new pharmacological properties such as anticancer, anti stroke and anti-angiogenic effects are introduced. There is no scientific report on noscapine effect on reproductive system. The aim of present study was to determine in vitro effect of ...

Effect of human chorionic gonadotropin (hCG) on expression of vascular endothelial growth factor a (VEGF-a) in human mid-secretory endometrial cells in three-dimensional primary culture.

Several lines of evidence suggest that human uterine endometrial cells can bind human chorionic gonadotropin (hCG) which, in turn, influences the physiology of implantation stage endometrium. Vascular endothelial growth factor (VEGF) appears to be a candidate mediator in this process. However, our knowledge about hCG action on VEGF in human endometrial cells is very thin. In the present study, ...
